Previous study [see above, Flower 1973] showed that benzydamine inhibited the formation of PGF2« and PGD2 but it potentiated the synthesis of PGE2 by approximately 150%.
All these experiments were performed in the presence of 1 mM substrate, concentration chosen since it gave an almost equal production of PGE and PGF and smaller amounts of PGD.
However, when the reaction was performed under optimal conditions for PGE formation (0.5 mM substrate) no potentiation was observed.
The IC50 for PGE formation was about 5 mM.
The shape of the product substrate curve for PGE suggests strong substrate or product inhibition and the action of benzydamine could be the prevention of this situation, thus permitting an increase in the amount of PGE formed.
