"pre-powerstroke")

How myosin motors power cellular functions

Motor cycle of myosin motors. Central panel: schematic representation of thefour-step cross-bridge cycle proposed by Lymn and Taylor (1971) . Starting from a nucleotide-free myosin strongly bound to actin (rigor), in step 1, the actin–myosin complex dissociates as a result of ATP binding to myosin. In step 2, the detached myosin undergoes an isomerization (recovery stroke) that swings its lever arm (repriming) and allows the active site to become competent for ATP hydrolysis. In step 3, the myosin with its hydrolysis products rebinds to the actin filament. This is the starting position for the powerstroke. In step 4, the cross-bridge releases the hydrolysis products (Pi and then ADP) and generates force displacing the actin filament (blue numbering). Outer panel: structural view of the actomyosin motor cycle. The scallop striated muscle myosin II S1 fragment is shown in different structural states along the motor cycle. (A) Rigor state. In a nucleotide free state, the myosin motor domain (grey spheres) is strongly bound to the actin-filament (blue arrow). The lever arm is composed of the converter subdomain (green) and the light chain binding helix with two light chains bound: ELC (red) and RLC (light violet). The Relay helix spanning the motor domain (dark blue helix) and controlling the converter orientation is straight. (B) Post-rigor state. ATP-binding to the myosin motor domain induces a conformational change in the actin-binding site that causes the myosin head to release from the actin filament, whereas the lever arm position remains mostly unchanged. Further conformational rearrangements may occur resulting in the detached myosin state, which represents an alternate, parallel detachment path that could increase the number of heads detached from the filament for myosin II. (C) Pre-powerstroke state. Next, a large conformational change in the myosin head occurs, which closes the nucleotide binding pocket that is necessary for ATP hydrolysis. This recovery stroke that occurs for myosin when detached from actin reprimes its lever arm. The inorganic phosphate and ADP are trapped in the active site in this state; the Relay helix adopts a kinked conformation (dark blue helix). In this state, the myosin head rebinds to the actin filament with rather weak affinity. The powerstroke starts from this weakly attached ‘top of powerstroke state’. A number of transitions occur on actin that promote the controlled release of the inorganic phosphate and the MgADP. Each transition allows an increase in the binding affinity between myosin and actin. Details about how the lever arm is controlled during this power-stroke are not known, although it allows myosin to produce force and allows the lever arm to swing back to the rigor position of the acto-myosin complex.
