Lo scopo principale di questo emendamento è quello di modificare gli obiettivi primari e gli obiettivi secondari principali di efficacia nello studio Sarizotan/001/II/2015 (STARS). Nello specifico, l’attuale obiettivo primario di efficacia dello studio STARS, ovvero valutare l’effetto di sarizotan (da 2 a 10 mg BID) rispetto al placebo nella riduzione del numero di episodi di apnea, durante il tempo di veglia, in pazienti affetti dalla sindrome di Rett (RTT) con anomalie respiratorie, viene modificato per valutare l’effetto di sarizotan (da 2 a 10 mg BID) rispetto al placebo, sul carico del caregiver, basato sulla valutazione delle 3 preoccupazioni principali del caregiver (scala analogica visiva, [VAS]).
L’attuale obiettivo secondario principale di efficacia, ovvero valutare l’effetto di sarizotan (da 2 a 10 mg BID) rispetto al placebo sull’impressione del cambiamento valutata dal caregiver (CIC) dal basale, viene modificato per valutare l’effetto di sarizotan (da 2 a 10 mg BID) rispetto al placebo, sulla variazione globale dal basale, valutata tramite l’impressione clinica globale sul cambiamento (CGI-C) che viene esaminata dallo sperimentatore.
Come conseguenza di queste modifiche, i precedenti obiettivi primari e secondari principali di efficacia diventeranno obiettivi secondari. Inoltre, è stato modificato l’ordine di priorità per le analisi degli endpoint secondari di efficacia.
